腹腔注射卵清白蛋白致大鼠内脏高敏感的研究

摘　　要：目的 :建立鸡卵清白蛋白致内脏高敏感大鼠模型 ,研究该模型内脏高敏感与肥大细胞的关联。 方法 :腹腔注射鸡卵清白蛋白使大鼠内脏致敏 ,分别在给药 3d及 2周后用特殊染色法观察结肠肥大细胞的形态学改变。用腹部撤离反射 (abdom -inal withdrawal reflex,AWR)评估致敏大鼠对直肠扩张刺激的内脏感觉改变。 结果 :甲苯胺蓝染色法显示 ,致敏大鼠肠黏膜及肠系膜肥大细胞数量明显增加 (P<0 .0 1)。阿尔辛蓝 -藏红染色法显示 ,对照组及致敏给药后 3d的肠系膜肥大细胞内主要为未成熟颗粒 ,而致敏后 2周肥大细胞内主要为成熟颗粒。直肠扩张显示内脏致敏大鼠 3m l及 5 m l气囊组 AWR评分显著高于对照组 (P<0 .0 1)。结论 :腹腔注射鸡卵清白蛋白致内脏高敏感的作用确切 ,内脏致敏作用很可能和肥大细胞的增生和脱颗粒状态相关。Objective: To investigate the visceral hypersensitivity model induced by chicken egg albumin and the functional alteration of mast cells in this model. Methods: SD rats were sensitized by intraperitoneal injection of chicken egg albumin,the colon mast cells were stained to study the alteration of mast cells morphology.The responses of viscerosensitive neurons were recorded by the abdominal withdrawal reflex to investigate visceral nociceptive transmission on the repeated colon distention. Results:It was showed by toluidine blue stain that the number of mucosal and mesenteric mast cells increased significantly compared with the control group, the granules in mesenteric mast cells were immature in control group and the experiment group after 3 d, and it became mostly mature after 2 weeks in mesenteric mast cells by alcian blue Safranine staining.The result of AWR score among 3 ml and 5 ml distention groups increased significantly compared with control group( P <0.01). Conclusion: It is confirmed that intraperitoneal injection of chicken egg albumin can mediate colon hypersensitivity reaction.The results also suggest that the activated mast cells play an important role in the visceral hypersensitivity.
