摘  要:目的 :癌基因的激活或抑癌基因失活与肿瘤的发生密切相关 ,分析这些基因突变可能提供关于肺癌发病的病理机制的线索。方法 :对非小细胞肺癌 (肺鳞癌和肺腺癌 ,NSCLC)病人的肿瘤组织进行了抑癌基因p5 3、S期细胞比例 (SPF)、DNA含量等的研究 (免疫组织化学法 ,流式细胞术 ) ,结合病人的临床病理资料进行分析比较。结果 :肺鳞癌患者的吸烟指数较肺腺癌患者明显为高 (15 4 6 4 7± 795 0 7,815 0 0± 5 39 79,P <0 0 5 ) ,细胞增殖动力学参数S期细胞比例也较肺腺癌明显增高 (15 13± 4 4 6 ,7 4 9± 3 19,P <0 0 5 )。但p5 3过度表达、DNA倍体在 2种组织学类型肺癌之间无显著差异。结论 :肺鳞癌与吸烟关系较肺腺癌密切 ,且肺鳞癌具有更多的进展潜能。吸烟指数、p5 3过度表达及高SPF可作为肺癌病人预后不良的指征 ,但吸烟指数、p5Objective: Smoking indexes (BI) and some biological parameters were studied among 14 cases of squamocellular carcinoma and 20 cases of adenocarcinoma of the lung to analyze possible relationship occurring between smoking and those parameters in each histological type. Methods: The expression of p53 was tested with immunohistochemical technique; Cell cycle, DNA ploidy, and S phase fraction (SPF) were analyzed with flow cytometry. Results: It was found that BI and SPF were significantly higher in squamocellular carcinoma group than in adenocarcinoma group; while there was no difference of p53 expression in these 2 histological types. These data indicated that squamocellular carcinoma was more related to smoking behavior and had stronger evolution tendency. Conclusion: BI, SPF, and p53 overexpression might be used as biomarkers for prognosis of lung cancer, but not an independent biomarker for squamocellular carcinoma or adenocarcinoma in lung.
