ATRA对原代APL细胞分泌细胞因子影响的研究

摘　　要：目的 :探讨 APL 细胞经过 ATRA诱导后细胞因子分泌水平的变化。方法 :APL 患者的骨髓和外周血标本被收集于肝素抗凝管中 ,采用传统 Ficoll密度梯度法分离 APL 细胞。按照 FAB细胞形态学和细胞遗传学进行 APL 诊断。采用 EL ISA法测定原代白血病细胞培养悬液中的 IL- 1β、IL- 6、IL- 8、TNFα和 G- CSF含量。NBT还原实验检测 APL 细胞分化状态。结果 :APL 细胞在 ATRA10 - 6 m ol/L体外作用 96 h,或体内诱导作用 96 h,IL - 1β(P<0 .0 5 )和 G- CSF(P<0 .0 5 )分泌水平增加 ,IL - 6和 IL - 8水平降低 (P<0 .0 5 ) ,但 TNFα水平改变不明显。另外 ,体外 APL细胞的增生率与IL - 1β分泌率和 G- CSF呈正相关 ,检测到 IL - 1β或 G- CSF的患者的细胞数明显高于检测不到者。结论 :IL - 1β和 G- CSF在 ATRA作用后的Objective:To detect the modulation of production of hematopoietic growth factors by APL cells after incubation with exposure to ATRA.Methods:Bone marrow or blood samples from APL patients were collected in heparinized microfuge tube. Diagnoses were performed according to the FAB cytological classification criteria and cytogenetic criteria. Primary APL cells were separated and purified by traditional Ficoll Hypaque density centrifugation and enriched after adherence to plastic surfaces. IL 1β, IL 6, IL 8, TNFα and G CSF levels in the supernatants of cultured leukemia cell were estimated by ELISA method . NBT method was used to detect the differentiation of APL cells at the same time as evaluating hematopoietic growth factors . Results:96 H After exposure to ATRA 10 -6 mol/L in vitro of 60 mg/day in vivo,APL cells showed a significant increase of IL 1β( P <0.05) and G CSF ( P <0.05) production , and a significant decrease of IL 6( P <0.05 )and IL 8( P <0.05),however,there was no obvious variation of TNFα,if compared with that of APL cells without exposure to ATRA.On the other hand,the proliferation ratio of APL cells in vitro was statistically correlated to the IL 1β secretion ratio or G CSF secretion ratio.And the cell number ratio in patients with detectable IL 1β or G CSF was higher than that without detectable IL 1β or G CSF.Conclusion:IL 1β and G CSF secretion play an important possible role in the proliferation of APL cells after exposure to ATRA.
