摘 要:目的 :研究 1 92 Ir插植术治疗口腔鳞癌前后肿瘤组织 DNA含量的变化。方法 :选择 10例未经治疗的口腔癌患者 ,取放疗前新鲜肿瘤标本和放疗后 3天~ 7天手术新鲜肿瘤标本 ,通过流式细胞仪 (FCM)测定 DNA含量的 3个主要参数 ,异倍体 (aneuploid)、DNA指数 (DNA Index,DI)和 S期细胞比例 (S- phase fraction,SPF) ,比较显效组与非显效组、放疗前后 DNA含量之间的变化。结果 :显效组与非显效组的 DI分别为 1.6 3± 0 .10和 1.33± 0 .2 4(P<0 .0 5 ) ,SPF分别为 16 .6 1± 0 .86和12 .32± 4.2 0 (P<0 .0 5 ) ;放疗前后肿瘤组织 DI分别为 1.46± 0 .12和 1.2 0± 0 .11(P<0 .0 1) ,SPF分别为 14.0 3± 3.85和 8.0 8± 1.2 7(P<0 .0 1)。结论 :1 92 Ir插植术是治疗口腔鳞癌的有效方法 ,对口腔鳞癌 DNA含量的检测可以衡量放射治疗的效果并有助于制定治疗计划。Objective:To study the change of DNA contents before and after interstitial 192Ir brachytherapy in oral squamous cell carcinoma.Methods:Fresh specimens from 10 previously untreated patients with oral squamous cell carcinoma were selected before and after radiotherapy.Aneuploid, DNA index(DI)and S-phase fraction(SPF)were measured by flow cytometry(FCM).These parameters were compared.Results:DI of obvious effect group and un-obvious effect group were 1.63±0.10 and 1.33±0.24, respectively(P<0.05). SPF of obvious effect group and un-obvious effect group were 16.61±0.86 and 12.32±4.20,respectively(P<0.05). DI of tumors before and after radiotherapy were 1.46±0.12 and 1.20±0.11,respectively(P<0.01). SPF of tumors before and after radiotherapy were 14.03±3.85 and 8.08±1.27,respectively(P<0.01).Conclusion:Interstitial 192Ir brachytherapy is a useful technique for oral squamous cell carcinoma. DNA measurements on oral squamous cell carcinoma can weigh effect of radiotherapy,and are helpful for planning an effective treatment.
