摘 要:目的 通过观察缺氧过程中大鼠脑皮质线粒体细胞色素氧化酶 (COX)活性、亚基 COX 、 的蛋白量的变化 ,探讨 COX活性改变及其调节的可能机制。 方法 成年雄性 Wistar系大鼠随机分为平原对照组、缺氧 2、5、15和 3 0 d组。缺氧大鼠于低压舱内模拟海拔 5 0 0 0 m高原连续减压。氧电极法测量脑皮质线粒体 COX活性 ,Western blot分析线粒体 COX 、 蛋白。 结果 缺氧 15 d内 ,COX活性持续下降 ,与对照组差异有显著性意义 ;缺氧 3 0 d时 ,COX活性较缺氧 15 d时显著升高 ,但仍低于平原水平 ,差异有显著性意义。各组线粒体 COX 、 蛋白量差异无显著性意义。 结论 缺氧可影响脑皮质线粒体 COX活性 ,这可能是缺氧导致线粒体氧化磷酸化功能改变的原因之一 ;COX 、Objective To explore the changes and regulation of cytochrome oxidase(COX) activity in cerebral cortex mitochondria of rats after prolonged hypoxia. Methods Wistar Rats were randomly divided into control and 4 experimental groups. The rats of 4 experimental groups underwent prolonged hypoxia in a hypobaric chamber simulating 5 000 m high altitude for 2, 5, 15 and 30 days respectively. Mitochondria were isolated from cerebral cortex by centrifugation. COX activity was measured by Clark oxygen electrode. The protein contents of COX subunits I and IV in mitochondria were detected by Western blot analysis. Results Up to 15th day hypoxic exposure, COX activity decreased continuously, down to 57% of the control level; but restored to 78% of the control level in 30 d group. The differences between protein contents of COX subunits I and IV in mitochondria from cerebral cortex of experimental groups and control group were nonsignificant. Conclusions The changes of COX activity may contribute to the disorder of mitochondrial respiratory function and energy pruduction during hypoxia; other mechanisms rather than the changes of COX subunit I and IV protein content participate in the regulation of COX activity.
