阿斯匹林对平阳霉素诱导培养兔晶体白内障形成抑制作用的实验研究

摘　　要：目的 :观察阿斯匹林对平阳霉素诱导培养兔晶体白内障形成的抑制作用 ,研究阿斯匹林对晶体上皮细胞凋亡的抑制作用。方法 :采用兔晶体体外培养方法 ,观察阿斯匹林对不同培养时间 (2 4h ,4 8h ,72h ,96h)平阳霉素诱导兔晶体混浊形成的影响。在透射电镜下观察晶体上皮细胞超微结构的改变。通过免疫组织化学S P染色检测不同培养时间 (12h ,2 4h ,36h)晶体上皮细胞Bcl 2表达的情况。结果 :阿斯匹林可延缓和减轻平阳霉素诱导的兔晶体混浊的形成。培养 2 4h透射电镜下平阳霉素组和平阳霉素 +阿斯匹林组均检测到凋亡细胞。阿斯匹林 +平阳霉素组和平阳霉素组Bcl- 2染色的积分光密度在 12h分别为 2 5 1.0 1± 75 .2 7和 12 2 .2 3± 4 0 .74 (P <0 .0 5 ) ,在 2 4h分别为 178.6 3± 16 .84和 96 .15± 15 .76 (P <0 .0 5 ) ,在 36h分别为 114 .14± 2 9.86和 6 4 .71± 15 .5 3(P <0 .0 5 )。结论 :阿斯匹林可抑制平阳霉素诱发的兔晶体上皮细胞凋亡 ,延缓和减轻平阳霉素诱导的兔白内障的发生和发展。Objective: To observe the inhibitory effect of Aspirin on cataractous lenses of rabbits induced by pingyangmycin.Methods:The rabbits lenses were cultured in vitro and cataractous lenses were induced by pingyangmycin.After treated with Aspirin, the effect of the drug was observed at different time points(24 h,48 h,72 h,96 h ).Results:Under transmission electron microscopy,the occurrence of programmed cell death were measured in the pingyangmycin group and pingyangmycin+Aspirin group.The integral optical desity values for expression of bcl 2 were enhanced in pingyangmycin+Aspirin group( P <0.05).Conclusion:Aspirin has an inhibitory role in pingyangmycin induced rabbit lens epithelial cell apoptosis and can delay and ameliorate the occurrence and development of cataract.
