摘  要:目的 观察中枢给予外源性睫状神经营养因子 (CNTF)对抑郁大鼠行为和海马神经元损伤的影响。方法 将 60只SD雄性大鼠随机分为正常对照组 (NC组 )、生理盐水 +抑郁组 (S +D组 )、CNTF 3 0 0 0U +抑郁组 (Ch+D组 )和CNTF 30 0U +抑郁组 (Cl+D组 ) ,每组 1 5只。采用长期不可预见性中等强度应激造成大鼠抑郁模型 ,于实验开始前 1天和实验第 7,1 4 ,2 2天用Openfield法测定大鼠行为 ,于应激第 2 2天将其处死 ,固定脑组织 ,用组织化学、光镜和电镜等技术观察海马CA1、CA3及齿状回神经元的数目和形态。结果  (1 )应激第 2 2天 ,S +D组体重增长的g数 [(2 8 2 2± 4 2 2 )g]和蔗糖溶液消耗量 [(7 33± 2 43)g]均低于NC组 [分别为 (1 2 4 95± 6 44)g和 (2 0 50± 4 80 )g;P <0 0 1 ] ;而Ch+D组 [分别为 (64 72± 1 5 69)g和 (1 5 56± 3 48)g]均高于S +D组 (P <0 0 1 )。 (2 )Ch+D组的抑郁行为 [水平运动为 (9 80± 1 2 3)次 / 3min ,垂直运动中位数为 3次 / 3min]较S +D组 [水平运动为(1 1 6± 0 41 )次 / 3min ,垂直运动中位数为 0次 / 3min]有显著改善 (P <0 0 1 )。 (3)S +D组和Cl+D组各脑区神经元的丢失均多于NC组 (P <0 0 5和P <0 0 1 ) ,而Ch+D组神经元的丢失则少于S +D组和Cl+D组 (P <0Objective To observe the effects of intracerebroventricular ciliary neurotrophic factor (CNTF) on the depressive behavior and hippocampal neurons in the depression model rats Methods Sixty SD male rats were divided into four groups One group was given no treatment as control (NC) The rats in other three groups were given chronic unpredictable mild stress to establish depression model, and intracerebroventricular microinjection saline (S+D), CNTF 3 000 U (C h+D) or CNTF 300 U (C l+D), respectively Drugs of different dosage were intracerebroventricular microinjected to all the depressed groups on the 1, 3, 5, 7, 9, 11 and 13th day, just before stress beginning Openfield test was used to observe the behavior of rats on the day before stimulation, and the 7, 14, 22nd day during experiment On the 22nd day, animals were put to death and the brains fixed The morphological and number changes of hippocampal neurons in the depressive rats were determined with methods of histochemistry, light microscope and electron microscope Results (1) On the 22nd day, the increase of body weight [(28 22±4 22)g] and consumption of saccharose [(7 33±2 43)g] in S+D group were lower than that in NC group [(124 95±6 44)g and (20 50±4 80)g, respectively; P <0 01]; while both were higher in C h+D group [(64 72±15 69)g and (15 56±3 48)g, respectively] than in S+D group ( P < 0 01) (2) The depressive behavior in C h+D group [crossing: (9 80±1 23)times/3 min, rearing: median 3 times/3 min] was significantly improved compared with the S+D group [crossing: (1 16±0 41) times/3 min, rearing: median 0 times/3 min; P <0 01] (3) The neuron loss in S+D and C l+D group were both more severe than in NC group ( P <0 05 and P <0 01), while that in C h+D group was less severe than in S+D group and C l+D group ( P <0 05) (4) The activity of ACP in different brain areas of C h+D group was significantly decreased Conclusion CNTF might improve depressive behavior by protecting
