摘  要:目的 观察地塞米松调节人卵巢癌细胞系 3AO细胞增殖分化的作用机制及 3AO细胞中糖皮质激素受体 (GR)的表达。方法 ①采用氨基安替比林法测定糖皮质激素受体拮抗剂 (RU 4 86 )作用 3AO细胞后阻断糖皮质激素的效应 ;②采用放射配体结合分析法检测 3AO细胞中GR的表达 ;③采用定量RT PCR法检测地塞米松对 3AO细胞中GR的调节作用。结果 ①RU 4 86可完全阻断地塞米松对 3AO细胞活性的诱导作用 ;②3AO细胞中存在高亲和力、低容量糖皮质激素受体 ;③地塞米松使GR结合活性明显降低 ,且具有时间依赖性。结论  3AO细胞中存在GR ,地塞米松调节 3AO细胞增殖分化的作用由GR介导 ,地塞米松可向下调节 3AO细胞GR的结合活性 。Objective To study the effects of dexamethasone on the proliferation and differentiation of a human ovarian carcinoma cell line (3AO) and on the expression of glucocorticoid receptor(GR). Methods ①Alkaline phosphatase(AKP) activity was determined as described. ②The expression of glucocorticoid receptor in 3AO cells was studied through utilizing radioligand bingding assay. ③The regulation of glucocorticoid receptor in 3AO cells was further studied with quantitative RT PCR. Results The induction of AKP activity by dexamethasone was blunted by RU 486, a potent glucocorticoid antagonist. There existed high affinity and low capacity GR in 3AO cells, the GR binding activity could be down regulated by dexamethasone in a time dependent way. Conclusion Glucocorticoids play an important role in the regulation of 3AO cell proliferation and differentiation. There is presence of GR in 3AO cells and the cellular effects of dexamethasone on 3AO cells is mainly mediated via GR and GR can be down regulated by dexamathasone.
