摘  要:目的 研究持续压力对体外培养大鼠成骨细胞碱性磷酸酶分泌活性的影响。方法 利用自行研制的细胞加力装置给大鼠成骨细胞施加不同大小的静压力 ,观察加力不同时程后的碱性磷酸酶分泌活性。结果 加力后成骨细胞碱性磷酸酶分泌活性降低 ,停止加力后 2 4h碱性磷酸酶的分泌活性在 3× 10 5Pa组恢复正常 ,而在 4× 10 5Pa和 5× 10 5Pa组未完全恢复。结论 持续压力可降低成骨细胞碱性磷酸酶的分泌活性 ,适当力值的压力在恢复正常后碱性磷酸酶的分泌活性可恢复正常 ,力值过大则可能影响细胞的功能。Objective To investigate the effect of continuous stress on secretion of alkaline phosphatase(ALP) in cultured rat osteoblasts in vitro.Methods Static continuous stress of different magnitude were applied on cultured rat osteoblast through infliction apparatus devised by us; secretions of ALP were observed after different durations of application of stress.Results Decreased ALP secretion were observed after application of stress; in 3×10 5 Pa group, secretion level recovered to normal 24 h after stopping infliction; whereas in 4×10 5 Pa and 5×10 5 Pa group, the enzyme secretion did not recover.Conclusion Secretion of osteoblast ALP decreased after being applied continuous stress; it can restore to normal level after dismission of a moderate magnitude of stress. Cellular function can be damaged by overload on cells.
