雌激素受体在人舌鳞癌中的表达及β-雌二醇对其增殖的影响

摘　　要：目的 :观察雌激素受体 (ER)在人舌鳞癌Tca81 1 3细胞系细胞上的表达 ,研究 β 雌二醇 (estradiol,E2 )对培养的舌癌细胞增殖的影响。方法 :用免疫细胞化学方法和RT PCR方法检测雌激素受体在人舌鳞癌细胞上的表达 ,将β 雌二醇 (β E2 )作用于体外培养的人舌鳞癌细胞 ,测定3H TdR掺入量 ,并用流式细胞仪测定细胞周期。结果 :经免疫细胞化学和RT PCR方法可检测到ER α和ER β在人舌鳞癌细胞上均有表达 ,其中ER β表达相对较弱。不同浓度的 β E2 可增加3H TdR掺入量 ,并存在着剂量效应。 1 0 - 6 mol/L的 β E2 能使人舌鳞癌细胞处于G1 期的细胞比例从对照组的 76 .5 %下降至 62 .3 % ,而处于S期和G2 期的细胞比例从 2 3 .5 %上升至 37.7% ,细胞的DNA合成增加 ,促进细胞的增殖。β E2 对舌癌细胞的促增殖作用可被Tamoxifen阻断。结论 :在人舌鳞癌细胞上有雌激素受体的表达 ,且 β E2Aim:To observe the expression of estrogen receptors alpha and beta in human tongue squamous cancer line Tca8113 cell, and to study the influence of β estradiol (β E 2 ) on the proliferation and cell cycle of cultured Tca8113 cell. Methods:Immunocytochemistry and RT PCR methods were used to observe the expression of estrogen receptors (ER) in human tongue squamous carcinoma line Tca8113 cell. 3 H TdR incorporation and cell cycle analysis were used to examine the change of proliferation and DNA synthesis of Tca8113 cell. Results:ER α and ER β mRNA were expressed in human tongue squamous cancer cell, and the expression of ER β was weaker than that of ER α.β estradiol at 10 -8 mol/L～10 -6 mol/L could increase the proliferation of human tongue squamous carcinoma cell in a dose dependent manner ( P <0.01).β E 2(10 -6 mol/L) could increase the proportion of cells in S phase and G 2 phase from 23.5% up to 37.7%. The effect of estradiol on the proliferation of cultured human tongue squamous cancer line Tca8113 cell could be inhibited by Tamoxifen. Conclusion:There are ER α and ER β expression in human tongue squamous cancer line Tca8113 cell, and β estradiol promotes the proliferation and cell cycle of cultured human Tca8113 cell.
