摘　　要：目的　探讨流式细胞术 (flowcytometry ,FCM)对诊断血小板无力症 (Glanzmann′sThrombasthenia ,GT)的意义。方法　采用流式细胞术及Western印迹分别检测正常人、GT患者及其父母血小板膜GPⅡb和GPⅢa的表达。结果　与正常人相比 ,GT患者GPⅡbⅢa明显减少 ,其父GPⅢa亦减少 (P <0 0 1) ,其母GPⅢa无差异性 (P >0 5 )。结论　FCM是诊断GT的一种简单、灵敏而可靠的方法 ,而且它还可用于GT的家系分析。Objective To investigate the significance of flow cytometry in the diagnosis of Glanzmann′s Thrombasthenia (GT).Methods Expression of platelet membrane GPⅡb and GPⅢa of the patients with GT and their parents was determined by flow cytometry, Western bloting.Results In the patient with GT, GPⅡb and GPⅢa expression was remarkably lower than that of normal subjects(P<0 001), In his father, GPⅡbⅢa was also relatively decreased(0 01>P>0 005). In his mother, results were not different from that of normal subjects(P>0 5).Conclusions The measurement of platelet membrane GPⅡbⅢa complexes by FCM can provide a simple, sensitive and reliable method for G T diagnosis,and also can be applied to analysis of GT family pedigree.
