摘  要:目的 :研究中药脑宁康 (NNK)对脑缺血预适应 (cerebralischemicpreconditioning ,CIPC)模型大鼠不同时刻ET 1、NO含量的影响 ,以观察中药对缺血脑组织的保护作用。方法 :健康雄性成年Wistar大鼠 114只 ,随机分为假手术组 (FO )、脑缺血预适应 (CIPC)模型组、缺血再灌注 (is chemicreperfusion ,IR)模型组、NNK 1组、NNK 2组、ASP(阿司匹林 ) 1组、ASP 2组。除FO组外 ,每组再分为缺血再灌注后 8,2 4 ,72h 3小组 ,总计 19组 ,每组 6只 ,复制CIPC模型和IR模型。观察在IR后不同时刻大鼠血及脑组织ET 1、NO含量的变化。结果 :IR后 8,2 4 ,72h各时刻血、脑组织ET 1及脑组织NO含量均有不同程度升高 ,血清NO含量均有不同程度下降 ;CIPC模型组血、脑组织的ET 1及脑组织的NO含量降低 ,血浆NO含量升高。结论 :NNK通过调控血及脑组织的ET 1、NO而扮演药物预适应角色 ,具有模拟或强化CIPC的作用。Objective:To research the influence of Naoningkang (NNK) on endothelin 1 (ET 1) and nitric oxide (NO) level in the model rats of cerebral ischemic preconditioning (CIPC), and observe the protective effects of herbs on ischemic cerebral tissue. Method:114 healthy male Wistar rats were randomly divided into 7 groups,i.e. false operation control (FO)group, cerebral ischemic preconditioning (CIPC) group, ischemic reperfusion (IR) group, NNK 1 group, NNK 2 group, ASP 1 group and ASP 2 group. Each group was divided into 3 groups except FO group,i.e. 8 hours, 24 hours and 72 hours after ischemic reperfusion. There were 19 groups in total. Each group included 6 rats. The models of cerebral ischemic preconditioning and ischemic reperfusion were duplicated respectively. The level of ET 1, NO in blood and cerebral tissue at different intervals were assayed. Result: After IR, ET 1 in blood and cerebral tissue and NO in cerebral tissue at different intervals all increased to different degrees, and NO in blood decreased to different degrees; CIPC accomplished protecting function on ischemic cerebral tissue by reducing ET 1 in blood and cerebral tissue and NO in cerebral tissue, as well as rising NO in blood. Conclusion: NNK serves as pharmic preconditioning role by regulating ET 1 and NO in blood and cerebral tissue. NNK has the function of simulating or strengthening CIPC.
