摘  要:目的 :观察和探讨非放射性核素 溴脱氧尿嘧啶核苷 (BrdU)标记在口腔肿瘤增殖研究中的价值。方法 :用 80 μg/ml的BrdU体外掺入舌癌细胞或舌癌组织 ,标本连续切片与增殖细胞核抗原 (PCNA)表达比较 ;相同孵育时间的细胞与流式细胞术 (FCM)定量S期细胞数比较。结果 :BrdU标记细胞核大浑圆 ,呈散在或姊妹细胞分布 ,子代细胞染色稍淡。BrdU掺入和PCNA表达之间无显著性差异 (P >0 0 5 )。FCM测定的S期细胞数显著多于BrdU掺入的细胞数 (P <0 0 0 1)。结论 :BrdU标记可用于口腔肿瘤细胞增殖研究 ,其应用潜力较大。Objective: The aim of this study is to evaluate bromodeoxyuridine (BrdU) label in the proliferation of the tongue cancer.Methods: The authors used 80 μg/ml BrdU to label three cases of fresh tissue of tongue cancer and four bottles of Tca 8113 cells in logarithmic growth period in vitro.They were fixed and embedded in the routine procedure,and then in serial sections.These specimens were labelled with BrdU polyclonal antibody (1∶200),and PCNA monoclonal antibody (1∶100),stained using immunohistochemical ABC method.The authors observed the shape and size of those positive cells,counted their label indexes (LI) of BrdU positive cells respectively.Four bottles of Tca 8113 cells were cultured for 1~4 days.One bottle of the cells was every day used in the exprerimental study about comparing the cells labelled BrdU with those of the S phase of FCM in same cultural time.Results: The positive cells generally owned a larger round nucleus,sometimes in company with sister cells.There was no difference of positive cells in these two kinds of labelling methods between the incorporation of BrdU and the expression of proliferation cell unclear antigen ( P >0 05).The cells of S phase with FCM method were much more than that method with BrdU incorporation ( P <0 001).Conclusion: The method of BrdU labelling could be used in studies of oral tumor cellular proliferation.
