运动对大鼠慢性颞叶癫痫模型痫性发作和脑内强啡肽A_(1-17)含量及分布的影响

摘　　要：目的　探讨有氧运动对痫性活动的影响状况及其可能的机制。方法　 Wistar大鼠经匹罗卡品化学点燃成功 45 d后 ,将存活鼠 (致痫鼠 )随机分为运动组和安静组。分别监测两组动物在有氧运动中及随后 1h这一时间段内其痫性发作频次。运动阶段 (45 d)结束后 ,应用免疫细胞化学的方法研究运动对内源性抗痫物质—强啡肽A1 - 1 7含量及分布的影响。结果　两组之间平均痫性发作频次对比 ,无显著差异 (P>0 .0 5 )。运动组较安静组在海马结构、内嗅皮质、扣带回及额叶眶部等皮质脑区中 ,强啡肽 A1 - 1 7的密度和免疫反应强度显著增加 (P<0 .0 0 1)。结论　运动并不能被看作一种癫痫发作的诱发因素。脑内强啡肽 A1 - 1 7在运动中释放增多 ,其神经元的神经化学可塑性变化可能体现了内阿片肽强啡肽 A1 - 1 7促使兴奋Objective To clarify better the relationship between exercise and epilepsy,an animal model of temporal lobe epilepsy(the pilocarpine model of epilepsy) was used. Methods The epileptic rats were divided into two groups during the chronic phase. These animals were continuously monitored during the periods of aerobic exercise program and an subsequent hour for 45 days,then we observed the contents and distribution changes by immunocytochemical identification of endogeous dynorphin A 1 17 immunoreactivity in brain. Results The mean frequency of seizures in the training group and silent group did not change significantly ( P >0.05). It was found that dynorphin A 1 17 immunoreactive neurons were significantly increased in the hippocampal formation and other cortical regions projected topographically by the efferent fibers of hippocampus after aerobic physical exercise program ( P < 0.001 ). Conclusion Physical activity in general can not be considered a seizure inducing factor. We propose that the neurochemical plasticity of these neurons may represent an attempt in which the endogenous opioid peptide dynorphin A 1 17 acts to restore the balance of excitatory/inhibitory neurotransmission.
