应用NMDA受体拮抗剂MK-801治疗局灶性脑缺血的实验研究

摘　　要：目的 :研究N 甲基 D 门冬氨酸 (NMDA)受体拮抗剂MK 80 1对大鼠局灶性脑缺血模型的治疗作用和治疗时窗 ,为临床治疗提供依据。方法 :采用线栓法阻塞大鼠右侧大脑中动脉制成局灶性脑缺血模型。以 [3H]噻吩环己哌啶 ([3H]TCP)为配基 ,采用宏观和微观放射自显影术观察不同时间缺血组和给药组NMDA受体通道的活性变化。结果 :缺血前及缺血 2h内给予MK 80 1(0 .8mg/kg体重 )可有效拮抗NMDA受体的过度激活。而缺血 3h后给药则无此作用。结论 :MK 80 1对局灶性脑缺血的治疗时窗在Objective:To study the therapeutic effects on ischemic penumbra and determine the therapy time window of MK 801,an antagonist of N methyl D aspartate(NMDA)receptor.Methods:Rat models with focal cerebral ischemia were made by the right middle cerebral artery occlusion. Autoradiography was used to examined the change of NMDA receptor activity with TCP as the binding ligand in brain tissue by intrathecal injection into the lateral ventricles.Results:It showed significant therapeutic efficacy of MK 801 in reducing the over excitation of NMDA receptor when given at 0.8mg/kg before ischemia or within 2 hours after ischemia, while it showed no efficacy when injected after 3 hours following focal cerebral vascular occlusion. Conclusion:MK 801 do have neuroprotective effects on ischemic penumbra and the therapy time window of MK 801 was within 3 hours.
