摘  要:用ELISA测IL 12、IFN γ水平 ;用原位杂交染色法测IL 12mRNA表达 ;电泳迁移率改变试验 (EM SA)测NF AT活性 结果表明 :葡萄膜炎患者PBMCIL 12、IFN γ水平和IL 12mRNA表达以及NF AT活性都显著高于正常对照和其他眼疾病患者 (P <0 .0 1) IL 12、IFN γ水平异常增高是葡萄膜炎患者Th1/Th2 功能紊乱、Th1细胞功能亢进主要原因之一 ;而在转录水平上NF AT活性异常增高可能与患者IFN γ基因转录增强密切相关.To detect the The levels of IFN-γand IL-12 and IL-12 gene expression and the activation of NF-AT in peripheral mononuclear cells (PBMC) in patients of uveitis and investigate their relationship on uveitis Methods The levels of IFN-γand IL-12 were detected by ELISA; the IL-12mRNA was detected by situ hybridization(ISH)respectively; the activity of NF-AT was detected by electrophoretic mobility shift assay(EMSA). Result: The levels of IFN-γ?IL-12 and IL-12 gene expression and the activation of NF-AT in PBMC from the patients of uveitis were significance higher than normal contrast and other patients of ophthalmopathy (P<0.01).Conclusion: The abnormal expression of IL-12?IFN-γ and NF-AT, which is suggested that may contribute to the pathogenesis of uveitis .These results prompt that the imbalance of Th 1/Th 2 cell might exist in patients with uveitis. The significance higher of the regulation by NF-AT at transcription level may be associated with reinforcement transcription of IFN-γ in uveitis.
