HHV-6感染在鼻咽癌发病中的意义

摘　　要：目的:探讨鼻咽癌组织中人疱疹病毒6型(HHV-6)在鼻咽癌(Nasopharyngealcarcinoma,NPC)发病中的意义。方法:PCR检测27例正常鼻咽组织、21例异型增生及42例鼻咽癌组织中HHV-6DNA和EB病毒(Epstein-barrvirus,EBV)DNA存在状况,用免疫组化检测36例NPC组织中EBVLMP1蛋白的表达及40例NPC组织中p53基因的表达。结果:异型增生和NPC组织中HHV-6DNA的阳性率分别为4.7%和26.2%,EBVDNA分别为66.6%和95.2%;正常鼻咽组织不存在HHV-6DNA,EBVDNA为25.9%。NPC组织中EBV-LMP1蛋白阳性率为47.2%,HHV-6阳性NPC中LMP1表达阳性率(81.8%)显著高于HHV-6阴性者(32.0%)。HHV-6阳性与阴性的NPC中p53表达的阳性率无显著性差异,但HHV-6阳性的NPC中p53表达强度显著性增高。结论:NPC组织中存在HHV-6的感染,与EBV-LMP1蛋白表达上调呈正相关。提示在NPC的发生中HHV-6可能直接参与和(或)通过上调EBV-LMP1的表达而起作用,并可能参与鼻咽癌发病中p53过表达的调控。Objective:To study on significance of human herpesvius6(HHV-6)infction and its role in pathogenesis of Nasopharyngeal carcinoma(NPC).Methods:1.PCR was used to am-plify the HHV-6and EBV DNA from the paraffin-embedded tissues of nomal nasopharynx,pre-cancerous nasopharynx and Nasopharyngeal carcinoma.2.EBV-LMP1gene was detected in36tis-sues of NPC and p53gene was detected in40tissues of NPC by immunohistochemistry (IHC).Results:1.HHV-6DNA was detected in4.7%of tissuese of precancerous nasopharynx,26.2%of NPC and none in the nomal nasopharynx.Within the same tissues,EBV DNA positive rate were66.6%,95.2%and25.9%.2.EBV-LMP1expression were detected in47.2%.LMP1positive rat of HHV-6 + NPC(81.1%)was significantly higher than HHV-6 - NPC.3.p53expression were detected in80%.p53positive rat of HHV-6 + NPC was no significantly higher than HHV-6 - NPC,but there was significantly difference in the intensity of expression of theirs.Conclusion:NPC tissues can be infected with both HHV-6and EBV,HHV-6infection is correlated with the expression of EBV-LMP1,suggesting that HHV-6plays a role in carcinogensis of NPC via upregulating the expression of EBV-LMP1.HHV-6may be correlated with the regulating of p53expression in NPC.
