摘  要:目的 克隆新的肝癌相关基因并探索其在肝癌发生发展中的作用机制。方法 采用mRNA差异显示技术 ,分析肝癌和癌旁组织差异表达基因谱 ,获得的差异表达基因片断作为探针 ,筛选胎盘cDNA文库 ,以获得全长cDNA序列。通过Northern印迹杂交的方法 ,分析所获得的新基因在 42例肝癌组织中的表达情况 ,并分析其临床和病理学意义。结果 克隆了一个新的肝癌相关基因 ,命名为HCCA3 ,该基因的全长cDNA 170 6bp ,编码一个 2 6 4个氨基酸的蛋白质 ,在人类肝癌中广泛表达 ( 78.6 % ,33/42 ) ,并与肝癌的浸润转移密切相关。结论 HCCA3是一个新的人类基因 ,在肝癌中的异常高表达可能对肝癌的发生发展具有重要的促进作用。Objective To clone novel hepatocellular carcinoma (HCC) associated genes that may provide new insight to molecular mechanisms of this disease development.Methods DD-PCR was employed to analyze altered gene expression patterns between HCC tissues and their adjacent non-tumor tissues.The cloned PCR fragment served as probe to screen a human placental cDNA library to obtain the full-length cDNA.mRNA expression level of the isolated gene were detected by Northern blotting in 42 patients with HCC and its clinopathological significance was analyzed.Results A novel HCC-associated gene named HCCA3 with a transcript of 1706 bp in length that encodes a protein of 264 amino acids was identified.Northern blot analysis revealed that HCCA3 mRNA was highly expressed in HCCs (78.6%,33/42) and significantly correlated with HCC metastasis.Conclusion HCCA3 is highly involved in HCC development.
