摘 要:目的 观察微管功能变化对C6胶质瘤细胞葡萄糖摄取的影响。方法 用影响微管功能的药物长春花碱 (抑制微管聚合 )、紫杉醇 (促进微管聚合 )与C6胶质瘤细胞在 37℃下孵育 3h后 ,再用含 12 5 μmol/L未标记 2 脱氧葡萄糖 ( 2 deoxy D glucose ,2 DOG)和 37kBq [3H] 2 DOG的磷酸盐缓冲溶液 (PBS) 1ml室温下孵育 5min。用液闪计数分析仪检测C6细胞葡萄糖摄取。GLUT1cDNA转染C6胶质瘤细胞后再与紫杉醇作用 ,观察转染后C6细胞的紫杉醇刺激葡萄糖摄取。结果 长春花碱抑制C6细胞葡萄糖摄取 2 4%~ 36 %。紫杉醇刺激葡萄糖摄取增加 16 %~2 3 % ;紫杉醇刺激GLUT1转染C6胶质瘤细胞的 2 DOG摄取增加约 35 %。结论 微管功能变化与C6细胞葡萄糖摄取及葡萄糖转运体介导的葡萄糖转运过程密切相关。Objective To investigate whether glucose uptake is mediated via the tubulin-microbubule system by C6 glioma cells.Methods Drugs that influence tubulin function,vinblastine (inhibiting tubulin polymerization) and paclitaxel (promoting tubulin polymerization),were used to incubate with C6 glioma cells for 3 h at 37 ℃,then the cells were incubated in 1 ml phosphate-buffered saline (PBS) containing 125 μmol/L unlabeled 2-deoxy-D-glucose (2-DOG),and 37 kBq [3H]2- deoxy-D-glucose for 5 min at 37 ℃.By using liquid scintillation analyzer,the glucose uptake by C6 glioma cells was determined.In the same way,the paclitaxel stimulated glucose uptake by GLUT1-transfected C6 cells was determined.Results Vinblastine inhibited basal glucose uptake by 24% to 36%.Glucose uptake was increased with paclitaxel by 16% to 23% and the paclitaxel stimulated glucose uptake by GLUT1-transfected C6 cells increased by about 35%.Conclusion The changes of microtubule function are involved in glucose uptake by C6 cells and glucose transport process mediated by GLUT1.
