椰毒伯克霍尔德氏菌——新发现的固氮菌及其简易鉴定方法

摘　　要：目的　验证椰毒霍尔德氏菌 (伯克氏菌 )的固氮能力并提出此菌的简易鉴定方法。方法　用无氮的无机盐基础培养基 (MBM) ,以葡萄糖为碳源 ,以微量接种此菌连续多次传代 ,与已知的非固氮菌和固氮菌进行比较。用 5项组合试验 (固氮、OF、脂粒染色、氧化酶、黄色素 )鉴定此菌。结果　全部 18株椰毒伯克氏菌能在无氮MBM上多次传代仍旺盛生长 ,比在营养丰富的培养基存活更长 ;5项试验 ,椰毒伯克氏菌呈独特的表型特征 ,与对照的 13株常见革兰氏阴性杆菌可以鉴别。结论　椰毒伯克氏菌为固氮菌 ,5项试验鉴定法可以准确简易地鉴定此菌。Objective To verify the nitrogen fixing capacity of Burkholderia cocovenenans and a simple identifying method.Methods Non nitrogen mineral basal medium (non N 2 MBM) with glucose as carbon source was inoculated with little amount of the bacterium.Nitrogen fixing positive and negative bacteria were also employed as control group at the same time.Five items of phenotypic characteristics (N 2 fixing,OF test,lipid granular stain,oxidase and yellow pigment) were tested with these bacteria.Results Burkholderia cocovenenans could grow and reproduce well on the non nitrogen MBM with glucose or other carbon source .Burkholderia cocovenenans strains presented species specific phenotypic characteristics in the 5 item group test.Conclusion Burkholderia cocovenenans is a N 2 fixing bacterium.The 5 item group test can be used as the simple and reliable identifying method for the bacterium.
