摘 要:目的 研究t PA在猪玻璃体腔注药联合睫状体冷冻诱导玻璃体后脱离的效果。比较动物实验中玻璃体后脱离 (PVD)的观察方法。方法 2 0只猪眼睫状体冷冻后 ,随机一眼注入t PA 5 0 μg ,另眼注射同体积的BSS溶液。注射后 6h和 12h进行A、B型超声检查 ,处死后眼球标本送光镜检查和电镜检查 ,观察PVD是否存在。结果 电镜结果实验组与对照组相比有统计学意义 (8∶1,P <0 .0 5 )。超声波检查结果 4∶1(6h) ,5∶1(12h)和光镜检查结果 (6∶1)与对照组相比无统计学差别 (P >0 .0 5 )。结论 在破坏血眼屏障后 ,玻璃体腔内注射t PA可以诱导PVD的发生 ;ObjectiveTo investigate the efficacy of intravitreal injection of Tissue Plasminogen Activator(t PA) combined with cyclocryoapplication on production of posterior vitreous detachment(PVD) in pig eyes and evaluate the methods of judging PVD in experimental animal study.MethodsTwenty eyes of 10 young pigs randomly received an intravitreal injection of t PA (50 μg/0.1ml) and the other one buffered salt solution (BSS) immediately after the cyclocryoapplication via conjuctiva. PVD were evaluated by ultrasonic examination (6 and 12 hours),light microscopy (12 hours),transmission electron microscopy (12 hours) and scanning electron microscopy (12 hours).ResultsThe eyes injected t PA combined with cyclocryoapplication exhibited a higher incidence of PVD compared to fellow eyes under the electron microscopy ( 8∶1, P < 0.05 ),and the results of ultrasonic examination (4∶1 for 6 hour,5∶1 for 12 hours) and light microscopy (6∶1)showed no significant difference between t PA group and BSS control ( P >0.05).ConclusionIntravitreal injection of t PA 50 μg combined with cyclocryoappliction can induce PVD in pigs.Transmission and scaning electron microscopy are more operative for the judgement of PVD than ultrasonic examination and light microscopy.
