摘　　要：目的　分析人胚眼视网膜色素上皮 (RPE)细胞的生物学特性。方法　酶分层消化法分离培养RPE细胞 ,原代细胞分别计算贴皿率、伸展率和生长率。传代细胞计算分裂时间和总分裂次数。结果　人胚RPE细胞贴皿率为85 7%± 5 2 % ,伸展率为 84 2 %± 4 8% ,生长率为 72 2 % ;成人RPE细胞的贴皿率为 79 4%± 8 3 % ,伸展率为80 3 %± 6 7% ,生长率为 63 8%。人胚RPE细胞的分裂时间 (DOT )为 1 5 8d ,总分裂次数 (PD)为 4 43 ;成人RPE细胞的分裂时间 1 91d ,总分裂次数为 3 66。ObjectiveTo analyze the biological characteristics of human fetal retina pigment epithelium(hfRPE).MethodsRPE cell of six embryonic eyeballs and four grown up eyeballs after corneal removing was isolated by enzyme digestion.Primary cells' plating rate,extension rate and growth rate were calculated.Subcultured cells were counted every day and growth curve was generated.Their doubling time(DOT) and population doubling(PD) were calculated.Cells were stained and identified with immunocytochemical anticytokeratin(AE1/AE3).Ultrastructure of cells were examined using scanning and transmission electron microscopy.ResultThe plating rate of hfRPE cells was 85 7%±5 2%,extension rate 84 2%±4 8% and growth rate 72 2%. The plating rate of adult RPE cells was 79 4%±8 3%,extension rate 80 3%±6 7%and growth rate was 63 8%.The DOT of hfRPE cells was 1 58 days and PD 4 43 days.That of adult RPE was 1 91 days and PD 3 66 days.Anticytokeratin staining was positive.ConclusionGrowth of hfRPE cells is a more active process than adult RPE cells in vitro.
