细胞周期蛋白cyclin D1、CDK4在口腔鳞状细胞癌中的表达及其意义

摘　　要：目的　分析口腔鳞状细胞癌细胞周期蛋白cyclinD1、CDK4的表达与临床病理资料及预后的关系。方法　回顾性研究 5 0例口腔鳞状细胞癌及 10例正常口腔粘膜cyclinD1和CDK4的表达情况 ,采用SABC法根据染色指标计数 ,并利用Cox比例风险模型进行生存多因素分析。结果　cyclinD1和CDK4在正常口腔粘膜上皮呈现低水平表达 ,在口腔鳞状细胞癌组织中过表达。单因素生存分析表明cyclinD1的表达与生存期显著相关 ,多因素Cox回归分析表明cyclinD1的表达可作为口腔鳞状细胞癌辅助性的预后指标 ,cyclinD1与CDK4的表达呈负相关。结论　cyclinD1和CDK4的过表达与口腔鳞状细胞癌的发生发展有密切关系 ,其中cyclinD1与肿瘤的预后密切相关 ,可作为辅助性的预后指标。ObjectiveTo study the relationship between the expression of cyclin D1、CDK4 and clinical pathological characteristics or prognosis of oral squamous cell carcinoma (OSCC).MethodsThe specimens included 50 cases of OSCC and 10 cases of normal oral mucosa.The expression of cyclin D1 and CDK4 was detected by immunohistochemical method.The relationship between expression of cyclin D1、CDK4 and clinicalpathological characteristics or survival was analysed by Spearman correlation test or log rank test,respectively,while univariate and multivariate prognostic analyses were performed by using Cox's regression model.ResultsAll the specimens of normal mucosa showed cyclin D1 and CDK4 low expression while the specimens of OSCC showed overexpression.Log rank test showed that the expression of cyclin D1 was negatively correlated with survial and Cox's regression indicated that cyclin D1 was a prognostic marker of OSCC.Overexpression of cyclin D1 and CDK4 showed siginificantly correllation.ConclusionOverexpression of cyclin D1 and CDK4 may involve in the carcinogenesis of OSCC and resulted in a poor prognosis.Assessment of cyclin D1 expression was useful as a prognostic factor for OSCC.
