摘  要:目的观察胃癌组织中COX-2的表达情况,探讨Hp-cagA菌株感染与胃癌COX-2表达之间的关系,为胃癌的早期预防提供有价值的实验和理论依据。方法采用流式细胞术,定量检测及分析32例胃癌组织及相应的癌旁正常胃组织中COX-2的表达水平,并通过PCR技术对胃癌组织中cagA基因的扩增产物进行检测。结果 32例胃癌组织中,有27例COX-2过表达,19例cagA阳性的胃癌组织中COX-2的表达水平明显高于13例cagA阴性的胃癌组织。结论 COX-2在胃癌组织中过度表达,cagA菌株感染可上调胃癌组织中COX-2的过度表达。可能存在cagA菌株感染之外的其它因素或机制调节COX-2的表达。因此,在根治幼感染的同时,应用COX-2特异性抑制剂可能是一种有效预防胃癌发生、发展的新路。Objective To observe the status for COX-2 expression in human gastric cancer, and explore the association between Hp cagA strain infection and the expression of COX-2 in human gastric cancer, To provide a valuable clue and basic data for early preventiongastric cancer. Methods To quantitatively determine and analysis the expression of COX-2 in 32 specimens of gastric cancer, and paired normal gastric tissues with Flow Cytometry, anddetect the cagA gene in gastric cancer by PCR. Results COX-2 is over-expression in 27 of 32(84%) specimens of gastric cancer, The expression of COX-2 in 19 specimens of gastriccancer with cagA positive strain infection is significantly stronger than 13 specimens with, cagA negative strain infection. Conclusion COX-2 is over-expression in gastric cancer and cagA positive strain infection could up-regulate over-expression of COX-2 in gastric cancer.There may be another way or channel to regulate the expression of COX-2 in gastric cancer besides cagA positive strain infection. Therefore, applying COX-2 selective inhibitors to prevent gastric cancer could be an effective and promising way.
