摘  要:目的 研究天津地区人群中葡萄糖激酶基因多态性与 2型糖尿病的关系。 方法 应用聚合酶链反应对天津地区的 4 3例 2型糖尿病患者与 4 6名正常对照个体的葡萄糖激酶基因的 3’末端 -10Kb处的 1个 (CA) n 重复片段的多态性进行了研究。 结果 在葡萄糖激酶基因中存在 5种等位基因(a、b、c、d、e等位基因 ) ,其中a等位基因频率在对照组 30 %、糖尿病组 16 % ,两组差异有显著意义 (P =0 .0 2 6 ) ,e等位基因频率在糖尿病组明显高于对照组 ,分别为 2 7%和 13% ,两组差异有非常显著意义 (P=0 .0 11)。在调整年龄、性别和体重指数后 ,e等位基因仍与 2型糖尿病存在特异性关系 (P <0 .0 5 )。 结论 葡萄糖激酶基因 (CA)n重复片段多态性可能是天津地区人群中Objective To study the association of the glucokinase gene polymorphism with type 2 diabetes in Chinese pupolation in Tianjin. Methods The (CA) n dinucleotide repeat polymorphism at locus 10 kb of 3' end of the glucokinase gene was detected by polymerase chain reaction in 43 type 2 diabetec cases and 46 normal controls. Results Five alleles were found in the glucokinase gene (a、b、c、d、e allele). The frequency of the a allele was 30% in case group and 16% in control group ( P =0.026). The e allelic frequency was significantly higher in the case group than in control group, 27% and 13% respectively ( P =0.011). After adjusting for age, sex, and body mass index, the e allele continued to have a significant association with type 2 diabetes ( P <0.05). Conclusion These results indicate that the dinucleotide (CA)n repeat polymorphism at the glucokinase locus is a genetic marker for type 2 diabetes in Chinese pupolation in Tianjin.
