天然细胞生长调控因子(NCGCF)对大鼠坐骨神经损伤后神经功能恢复的影响

摘  要:目的本研究是将天然细胞生长调控因子应用于周围神经损伤修复治疗的动物实验研究,试图在确定天然细胞生长调控因子促进损伤后坐骨神经功能指数恢复的同时探讨其可能的潜在机制。方法取65只重量为200~230g的雌性SD大鼠,随机分成3组:A组(假手术组)在术前和术后均不给药;B组为给药组,术前24小时和1小时分别皮下注射给药0.2ml,术后每天皮下给药0.2ml,术后连续给药1周;C组给同等剂量的生理盐水,其他同B组。在术后的1、4、10、14、21、28、35、42天进行足迹分析:PL:从足跟到足尖的最长距离,TS:第1趾到第5趾的连线距离,IT:第2趾到第4趾的连线距离,再根据Bain公式计算坐骨神经指数(SFI)。所有的数据使用SPSS统计软件进行独立样本t检验。结果坐骨神经功能指数的测定在A组(假手术组)未见明显的坐骨神经受损的表现。在B组和C组中,在术后的1、4天坐骨神经功能指数在-90~-100之间,两组之间并没有显著差异,在术后7,10、14、28天发现坐骨神经功能开始恢复,实验组效果要优于对照组(P<0.05),35天后两组之间没有显著差异。结论天然细胞生长调控因子能促进受损的坐骨神经功能指数的早期恢复其作用机制仍然不明确。ObjectiveOur research is designed to determine the therapeutic effect of NCGCF on repair of injured periph-eral nerve and explore the underlying mechanism.Methods 65adult female SD rats weighted200~230g were randomly divided into3groups.Group A(n=5)underwent a sham operation without crush and agent administration.In group B(n=30),each animal received subcutaneously injections of NCGCF0.2ml 24h and1h prior to nerve crush and then daily for the following7days.Group C (n=30)received injections of saline with the same protocol as group B.Motor functional assessment was performed on days1,4,7,10,14,21,28,35and42after nerve crushed in all rats.Animals were evaluated by a walking track test:The distance between the first and fifth toes(TS),the distance between the second and fourth toes(IT),and the print length(PL)were mea-sured.The sciatic functional index(SFI)was calculated and processed by SPPSS10.0using independent T test.Results There was no apparent functional deficit in the sham operated group,while in group B and group C animals showed almost complete sciatic dysfunction by SFI of-90~100in4days after nerve was crushed and there was no statistic difference between the2groups.From days14~28,NCGCF treated animals showed a statistically significant improvement over control group.After day28,no statistical difference was found between the two groups.ConclusionThe treatment of NCGCF is effective in promot-ing sciatic functional index recovery after nerve injury.The underlying mechanism is still uncertain and further investigation is expected.
