摘　　要：目的:探讨三七皂甙对百日咳菌液致大鼠脑水肿的治疗作用及机理。方法:SD大鼠随机分为生理盐水(NS)、百日咳菌液组(PB)、三七皂甙组(PNGS)、甘露醇组(MN)4组。采用百日咳菌液注入左颈内动脉导致大鼠脑水肿,测定左脑组织含水量(WC)、伊文思蓝(EB)、丙二醛(MDA)浓度、超氧化物歧化酶(SOD)活性、肿瘤坏死因子α(TNFα)水平,观察三七皂甙的保护作用。结果:左脑组织TNFα水平,PNGS组明显低于PB组和MN组(P<0.05)。左脑组织含水量、伊文思蓝含量:PB组较NS组明显增高,治疗各组均较PB组减少,且MN组>PNGS组(P<0.05)。治疗各组均能降低MDA浓度,提高SOD活性(P<0.05)。结论:三七皂甙对百日咳菌液致大鼠脑水肿具有治疗作用,其机制与抑制脂质过氧化和减少TNFα生成有关。Objective : To explore the mechanism and effects of panax notoginseng saponins (PNGS) on pertussis bacilli-induced brain edema in rats . Methods : The protective effect was studied of PNGS and mannitol (MN) on brain edema induced by injecting purtussis bacilli (PB) into the left internal carotid artery in rats . Forty Sprague- Dawley rats were randomly divided into four groups . Group 1 (NS , n = 10) , as control ;Group 2 (PB , n=10) , pertussis bacilli control ; Group 3 (PNGS , n=10) , PB + PNGS ; Group 4 (MN , n = 10) , PB + MN . The brain water content ( WC) , Evan's blue (EB) content , and the concentrations of malondialdhy (MDA) , superoxide dismutase (SOD) and TNFα were determined. Results : The levels of TNFα in left brain tissues of PNGS group were significantly lower than those of PB and MN groups ( P <0. 05) . WC ,and EB content and MDA concentrations in left brain tissues of all treat groups were significantly lower compared with that of the PB group , but SOD activity in left brain tissues of all treat groups were significantly higher than that of the PB group( P <0. 05) . WC , EB contents in left brain tissues of PNGS group were significantly lower than those of MN group (P<0. 05). Conclusion : The PNGS have a therapeutic effect on pertussis bacilli-induced brain edema in rats , and the effects are related to the inhibitory effect on lipid peroxidation , and decreased the release of TNFα.
