Study on the extracts of penicillium cyclopium Westling with reverse mutation assay

摘  要:从河南省林县食管癌高发区采集的500份陈粮中,分离鉴定出19株圆弧青霉,以Raulin—Thom氏液体培养基培养成膜状物和菌液,分别提取两种培养物,对所得各提取物进行细菌回复突变试验。结果2株提取物使TA98、TA100检试菌株呈现诱变阳性,16株提取物使E.coli ND—160产生回复突变阳性(84.21%)。表明圆弧青霉产毒菌株广泛存在,其提取物能使检试菌遗传基因DNA产生碱基对置换和移码突变。19 strains of P. cyclopium were isolated and identified from 500 grain samples of Linxian County, Henan Province, an area of high incidence of esophageal cancer. They were inoculated in the Raulin-Thom liquid medium and incubated at 25℃ for 12 days. The fungal membrane and the liquid of the strains were extracted and then the two kinds of extracts obtained in each strain of P. cycloium. Reverse mutation assay was done on each extract. 2 strain extracts were proved to be mutagenic in Ames test with TA98, TA100. 16 out of 19 strain extracts induced different reverse mutation of the Esherichia coli strain ND-160. The results show that the presence of the toxin-producing strain of P. cyclopium are widespread, and that they can lead to direct and indirect base-pair substitution and frameshift mutation. As P. cyclopium is one of the dominant fungi contaminating the grain of Lixian County, it is valuable to explore and study the relation between the fungus and esophageal cancer in the future.
