摘 要:报告了人胎盘型谷胱甘肽S-转移酶(GST-π)抗体的制备和应用。免疫沉淀和免疫组化显示,该抗体与日本Sato教授赠送的GST-π抗体具有相同的特异性和反应性。用免疫金银法检测了40例肺癌组织和配对肺组织对GST-π抗体反应的情况,肺组织呈弱阳性反应,19例鳞癌均为阳性,腺癌和小细胞癌的阳性率分别为89%(16/18)和33%(1/3),与所赠送抗体检测结果相比,差别无统计学意义(P>0.05)。免疫吸印法(Western blot)表明肺癌组织中GST-π水平明显高于正常肺组织。GST-π是非小细胞肺癌的一种标志酶,兔抗GST-π抗体能够较精确、敏感地用于检测组织中的酶蛋白。Glutathione s-transferase(GST-π) antibody of human placental form was prepared. Its immunological speciality and reactivity was identical with that presented by Professor Sato, which was demonstrated by immunoprecipitation and immunohistochemistry assay. The reaction to GST-π antibody in 40 cases of lung carcinomas and the matched lung tissues was studied with immuno-gold silver method. All of 19 squamous cell carcinomas were positive for GST-π. Adenocarcinomas and small cell carcinomas were found to stain positively with GST-π, the positive rates 89% (16/18) and 33% (1/3) respectively. The increase of GST-π level in lung tumor tissues was also identified by Western blot assay. The results indicate that GST-π may be a useful marker for nonsmall cell lung cancer, especially squamous cell carcinoma, and that GST-π antibody can be used to detect precisely and sensitively the GST-π in tissues.
