摘  要:目的 探讨γ干扰素受体α(interforon gamma receptor alpha IFN-γRα)及细胞间黏附分子I(intercelluar adhesion moleeule I.ICAM-1)在大鼠肝卵圆细胞增殖模型中的表达情况及其生物学意义。 方法 采用二乙酰氨基笏(2-acetaminofluorene AAF)加三分之二肝切建立大鼠肝卵圆细胞增殖的模型,并以大鼠单纯三分之二肝切后的再生肝纠织、单纯灌AAF4天后的肝组织及正常大鼠肝细胞作对照,采用RT-PCR检测IPN-γRα、ICAM-1及甲胎蛋白(alpha-fetoprotein AFP)mRNA在模型组及对照组肝组织中的表达情况。结果 AFP,IFN-γRα及ICAM-1mRNA 表达水平在模型组肝切后4天及9天明显上调,AFP在肝切后9天、IFN-γRa及ICAM-1在肝切后4天的表达均显著高于对照组(P<0.05),在肝切后20天复常。结论 AFN-γRα和ICAM-1mRNA的高表达与肝卵圆细胞的增殖及分化过程密切相关。Objective To investigate expression of IFN-γRα. ICAM-1 and meaning of this expression in the model of proliferating liver oval cells of rat. Methods The model of proliferating rat liver oval cells was constructed by administration of AAF(2-acetaminofluorene) by gavage combined with 2/3 hepatectomy, and the regenerating liver tissues after simple 2/3 hepateclomy, the liver tissues after four-day administration of AAF and the normal liver tissues were taken as control. The mRNA of interferon gamma receptor alpha subunit (IFN-γRα) , ICAM-1 and alpha fetal-protein( AFP) in model group and control group were detected by RT-PCR . Results The levels of IFN-γRα, ICAM-1 and AFP mRNA were significantly increased on day 4 and 9 after hepalectomy in model group, and the levels of AFP mRNA on day 9 after hepatectomy as well as that of IFN-yRa and ICAM-1 mRNA on day 4 after hepatectomy were significantly higher than that in the control group ( P < 0.05) . All of three factors restored to normal level on day 20 after hepatectomy. Conclusion The expression of IFN-γRα and ICAM-1 is significantly correlated with the proliferation and differentiation of liver oval cells of rat.
