摘　　要：目的 从人血浆组份Ⅲ中制备天然血管生成抑制素(angiostatin)。方法 采用Lysine Sepharose-4B亲合层析、Sephcryl S200层析,纯化人纤溶酶原,经弹性蛋白酶水解、Lysine Sepharose-4B亲合层析制备血管生成抑制素。结果 获得相对分子质量43 000和35 000两种蛋白。经鸡胚实验,显示出明显血管生成抑制活性。结论应用该工艺从血浆中提取血管生成抑制素是可行的。Objective To prepare natural angiostatin from blood product fraction Ⅲ .Methods Purify human plasminogen by Lysine Sepharose-4B affinity chromatography and Sephacryl S200 chromatography, then prepare anigostation by lysis with elastase and Lysine Sepharose-4B affinity chromatography. Results Two kinds of protein, with the relative molecular weights of 43000 and 35000 respectively, were obtained and showed significant activity of angiostatin. Conclusion The procedure is feasible to the preparation of angiostatin from plasma.
