摘  要:目的 :研究基因重组人干扰素α 2b(rhIFNα 2b)对人白血病细胞株K562端粒酶活性及相应增殖能力的影响 ,以探讨其抗肿瘤机制。方法 :以rhIFNα 2b处理K562细胞 ,用TRAP -ELISA法测定rhIFNα 2b作用前后细胞端粒酶活性 ,并用血球计数板直接计数法结合台盼蓝染色测定rhIFNα 2b对K562细胞增殖的影响。结果 :rhIFNα 2b下调K562细胞端粒酶活性并抑制其增殖。结论 :rhIFNαObjective:To investigate the downˉregulation of telomerase activity in human leukamia cell K562with interferonα.Methods:The human leukamia cell K562in culture was treated by interferonαˉ2b.By using telomerase repeat amplification protocol(TRAP)combined with ELISA,the telomerase activity in human leukamia cell K562before and after the treatment was detected.By using directed counting with blood cell numbered card,the effect of interferonαˉ2b on the proliferation of human leukamia cell K562was detected.Results:The interferonαˉ2b could obviously downˉregulate telomerase activity and inhibit the proliferation of human leukamia cell K562.Conclusion:The interferonαˉ2b can inhibit the telomerase activity in human leukamia cell K562,which maybe one of its antiˉcancer mechanisms.
