摘　　要：目的　研究中药玉竹提取物B (EB -PAOA)诱导人结肠癌CL - 1 87细胞凋亡的作用。方法　将体外培养的人结肠癌CL - 1 87细胞与不同浓度的EB -PAOA共育 ,通过倒置显微镜和透射电镜对活细胞及固定染色后的细胞进行了观察 ,确认有无凋亡细胞 ;通过流式细胞仪检测了CL - 1 87细胞周期的变化和凋亡率。结果　在倒置显微镜和透射电镜下 ,可见到大量具有典型特征的凋亡细胞 ;流式细胞仪DNA直方图上出现了凋亡峰 ,凋亡率呈时间—剂量依赖性。细胞周期分析结果显示 ,G0 G1 期细胞减少 ,S期细胞减少 ,G2 M期细胞增多。结论　EB -PAOA能够诱导人结肠癌CL - 1 87细胞凋亡 ,这可能是EBObjective To study the actions of the extract B of Polygonatum Odoratum (EB-PAOA) on the induction of the apoptosis in human colon cancer CL-187 cells. Methods Human colon cancer CL-187 cells were cultured in vitro with different concentration of EB-PAOA.Living cells and dead cells were observed and detected whether there were apoptotic cells with an inverted microscope and electronmicrograph.Apoptotic rate and cell cycles were then detected through a flow cytometer. Results It can be observed that large number of apoptotic cells had representative morphological characteristics under the inverted microscope and electronmicrograph.An apoptotic peak appeared in DNA histogram of the flow cytometer.The apoptotic rate of cell was of time and dose dependent.The result analysis of cell cycle indicated that the number of cells of G 0/G 1 and S period reduced,and the number of G 2/M period increased. Conclusions EB-PAOA can induce the apoptosis of CL-187 cells,which may be one of the mechanism of EB-PAOA's anti-cancer effect.
