噻唑蓝法测定免疫偶合物对肿瘤细胞的杀伤作用

摘　　要：目的 :探讨噻唑蓝法应用于测定免疫偶合物对肿瘤细胞的杀伤作用的可能性。方法 :将被测试物与肿瘤细胞共同孵育2h后撤去测试物 ,加入新鲜培养液培养48h ,然后加入噻唑蓝染液 ,4h后酶标仪测定。结果 :所测试的两种免疫偶合物对靶肿瘤有较强的细胞毒作用 ,这种作用明显强于对非靶细胞的作用。结论 :噻唑蓝法是一种敏感、简单、客观性强的测定免疫偶合物细胞毒作用的方法。所测定的两种免疫偶合物具有临床应用价值。Objective:To study the application of Tetrazolum dye in detecting the killing activities of the immunoconjugates on the tumor cells.Methods:The immunoconjugates were incubated with the tumor cells for 2 hours.Then the immunoconjugates were removed by washing and the cells were incubated continuously for 48 hours.After that,the Tetrazolum was added.4 hours later it was tested by Bio-tech.Results:The immunoconjugates shown optential specific cytotoxicities.The effect was stronger in targeted cells than that in non-targeted cells.Conclusions:The Tetrazolum dye is a sensitive,simple and objective way for detecting cytotoxicities of immunoconjugates.The immunoconjugates have a value of clinical application.
