端粒酶逆转录酶在口腔黏膜癌前病变和鳞癌中的表达

摘　　要：目的 探讨人端粒酶逆转录酶(human telomerase reverse transcriptase,hTERRT)mRNA在口腔癌前病变及鳞癌中的表达。方法 应用 MRNA原位杂交法对口腔鳞形细胞癌(oral sequamous cell carcinomas,SCC)22例,上皮异常增生20例,上皮单纯增生4例及正常口腔黏膜4例,进行了hTERT mRNA的检测。结果 正常口腔黏膜及上皮单纯增生组中hTERT mRNA表达较弱,阳性细胞主要位于基底层及棘细胞深层,阳性表达率为12.5％(1/8)。上皮异常增生组中hTERT mRNA的阳性细胞表达范围由基底层向多层上皮细胞扩展,表达率为45％(9/20)。SCC组中 hTERT mRNA阳性表达最强,表达率为 81.18％(18/22)。结论 hTERT的活化在口腔黏膜癌前病变癌变过程中起作用,随着细胞异型性的增加,hTERT mRNA阳性率及信号强度均增高。Objective To investigate the expression of telomerase hTERTmRNA in oral precancerous and sequamous cell carcinoma( SCC). Methods Expressions of telomerase hTERTmRNA in 22 cases of SCC, 20 cases of oral precancerous lesions(dysplasia lesion), 4 cases of hy-perplasia lesions and 4 cases of normal oral mucosa were examined. Results A weaker signal of hTERT mRNA expression was observed in normal oral mucosa and hyperplasia lesions. Positive cells were present in basal layer and hTERT mRNA positivity was 12. 5% (1/8). The positive cells of hTERT mRNA in the precancerous lesions extended from basal layer to multi-layered epithelium. The hTERT mRNA positivity was 45% (9/20). Strong hTERT mRNA expression was observed in SCC and its positivity was 81.18% (18/22). Conclusion The hTERT activity played an important role in the carcinogenesis of oral precancerous lesions. The hTERT positive rate was related to cell differentiation.
