摘  要:目的 探索氨肽酶抑制剂对体外培养人红白血病细胞株K5 62作用及其机制。方法 应用四甲基偶氮唑蓝 (MTT)法、荧光显微镜、透射电检测镜、流式细胞仪观察氨肽酶抑制剂对人红白血病细胞株K5 62生长的影响。结果 氨肽酶抑制剂能明显抑制白血病细胞生长 ,其半数生长抑制剂量为 2 8.8μg/ml;透射电镜超微结构证实氨肽酶抑制剂能诱发肿瘤细胞凋亡的典型形态变化 ;流式细胞仪检测证实氨肽酶抑制剂可诱发细胞凋亡 ;免疫组化SP法及流式细胞仪定量分析发现氨肽酶抑制剂能使癌基因Bcl 2表达降低、抑癌基因P53表达增强。结论 氨肽酶抑制剂能诱导K5 62细胞凋亡 ,其作用机制可能与癌基因Bcl 2表达减少。Objective To explore the function and mechanism of aminopeptidase inhibitor for extracorporeal cultivating human erythroleukemia cell line K562. Methods Methyl thiazolyl tetrazolivm method,fluorescence microscope, transmission electronic mirror and stream-cell meter were used to observe the influence of aminopeptidase inhibitor on upgrowth of human erythroleukemia cell line K562.Results Aminopeptidase inhibitor could markedly restrain leukemia cell hyperplasia with the half dose of upgrowth inhibitor 28.8 μg/ml, which validated in detail that aminopeptidase inhibitor could induce the typical configuration change in apoptosis of tumor cells by the ultrastructure of transmission electronic mirror; it could induce cell apoptosis by stream-cell meter and decrease cancer gene Bcl-2 expression and increase P 53 tumor suppressor gene by immunity group SP method and stream-cell meter through quantitative analysis. Conclusion Aminopeptidase inhibitor can induce the apotosis of K562 cell which bears relation to cancer gene Bcl-2 expression reduction and P 53 expression increase.
