摘 要:目的 构建高通量的组织微阵列 ,研究细胞增殖状态与卵巢肿瘤临床病理特征的关系 ,探讨组织芯片技术的可行性。方法 联合应用组织芯片和免疫组织化学技术 ,检测 2 90例卵巢上皮性肿瘤组织中Ki67抗原的表达情况 ,并以 13例正常卵巢组织作为对照。结果 卵巢癌组织Ki67指数显著高于卵巢上皮性良性和交界性肿瘤及正常卵巢组织 (P <0 .0 5 ) ;卵巢癌组织Ki67表达水平与卵巢癌组织学类型、细胞分化程度有关 (P <0 .0 5 ) ,而与患者年龄、FIGO分期无明显相关性 (P >0 .0 5 )。结论 Ki67能较好地反映卵巢癌细胞的增殖状态。组织芯片技术是 1种高效、快速。Objective To investigate the relationship of Ki67 with the clinicopathological characteristics of ovarian cancer by constructing and applying high-throughput tissue microarrays.Methods The expression of Ki67 was analyzed retrospectively by tissuemicroarray technology and immunohistochemistry in 290 patients with the epithelial ovarian tumors and in 13 control subjects with normal ovarian tissue.Results The Ki67 index was significantly higher in ovarian carcinoma than in benign tumors and normal ovarian tissues(P<0.05);The expression of Ki67 gene was closely related to histological classification and cancer cell differentiation(P<0.05).There was no significant relationship between Ki67 index and the age or FIGO staging of ovarian cancer patients(P> 0.05).Conclusion Ki67 is a favorable marker for the proliferation of ovarian cancer cells.Tissue microarray is an efficient and rapid technique of molecular biology.
