P-cadherin、PCNA和c-erbB-2基因表达与乳腺癌转移的关系

摘　　要：目的　探讨胎盘钙黏素 (P -cd)、增殖细胞核抗原 (PCNA )和c erbB 2基因表达与乳腺癌转移的关系。方法　采用逆转录聚合酶链反应 (RT -PCR )半定量的方法 ,测定 17例乳腺癌组织中P -cd基因的mRNA表达 ,采用免疫组化SP法测定PCNA和c erbB 2基因的蛋白表达。结果　无淋巴结转移组的P -cd校正值 ( 0 .9367± 0 .5 2 90 )明显高于有淋巴结转移组 ( 0 .35 88± 0 .12 85 ) ,有显著性差异 (t =3.0 2 8,P <0 .0 5 )。PCNA阳性表达率为 70 .6% ( 12 /17) ,其中无淋巴结转移组为 5 0 .0 % ( 4/8)、有淋巴结转移组为 88.9% ( 8/9) ;c erbB 2阳性表达率为 5 2 .9% ( 9/17) ,其中无淋巴结转移组为 2 5 .0 % ( 2 /8)、有淋巴结转移组为 77.8%( 7/9) ,两者无显著性差异 (P >0 .0 5 )。结论　PObjective To study the relationship of the expression of P-cadherin,PCNA and c-erbB-2 to breast cancer metastasis.Methods Reverse transcription-polymerase chain reaction(RT-PCR) was used to detect the expression of P-cadherin mRNA in 17 patients with breast cancer.S-P immunohistochemical technique was used to detect the protein expression of PCNA and c-erbB-2.Results P-cd was significantly higher in the axillary lymph node-negative patients than in the axillary lymph node-positive patients(0.9367±0.5290 vs.0.3588±0.1285,t=3.028,P<0.05).The positive expression rate of PCNA was 70.6%(12/17) for all patients,of which 88.9%(8/9) for patients with lymph node metastasis and 50.0%(4/8) for patients without.The positive expression rate of c-erbB-2 was 52.9%(9/17) for all patients,of which 77.8%(7/9) for patients with lymph node metastasis and 25.0%(2/8) for patients without.There was no significantly difference in the expression of PCNA and c-erbB-2 between the two groups(P>0.05).Conclusion Low P-cd expression is associated with axillary lymph node metastasis in breast cancer.
