p53、p21^(ras)蛋白和P-糖蛋白在胃癌中过表达的意义

摘　　要：目的 :研究胃癌组织p5 3、p2 1 ras蛋白和P 耐药糖蛋白 (P gp)在胃癌中的表达及临床意义。方法 :采用免疫组化SABC法检测 7例正常胃粘膜、1 2例慢性萎缩性胃炎、8例粘膜慢性胃炎伴肠型不典型增生和 93例胃腺癌组织中p5 3、p2 1 ras蛋白和P gp的表达。结果 :p5 3、p2 1 ras蛋白和P gp在正常胃粘膜组、癌前病变组和胃腺癌组中的阳性率分别为 0 ,1 4 3% ,0 ;2 0 0 % ,2 5 0 % ,5 0 % ;5 5 9% ,5 1 6 % ,4 4 1 %。p5 3蛋白、P gp表达均与淋巴结转移有关 (P <0 0 5 ) ;p2 1 ras蛋白表达与肿瘤的浸润程度有关 (P <0 0 5 ) ;P gp表达与p5 3和p2 1 ras表达有显著的协同性 (P <0 0 5 )。结论 :p5 3和p2 1 ras蛋白常并存于胃癌组织中。p5 3突变蛋白和P gp过表达在胃癌淋巴结转移中起重要作用 ,因此早期进行p5 3蛋白和PObjectives:To study the levels of p53,p21 \{ras\} protein and P glycoprotein(P gp) in tissues of gastric carcinoma tissues and its clinical significance.Methods:20 cases of gastric precancerous lesions and 93 adenocarcinomas and 7 normal control tissues were analyzed for p53,p21 \{ras\} and P gp by immunohistochemical staining.Results:The positive expressions of p53,p21 \{ras\} and P gp in normal,precancerous and adenocarcinomas were 0,14 3%,0;20 0%,25 0%,5 0%;and 55 9%,51 6%,44 1%,respectively.A significant correlation was found that both p53 and P gp overexpression were related to lymph node metastasis(P<0 05).p21 \{ras\} protein was also correlated to invasiveness(P<0 05),Conclusions:There are commonly co expression of p53 mutational protein,p21 \{ras\} protein and P gp in the tissues of gastric carcinoma.p53 and P gp overexpression may play an important role in the lymph node metastasis of gastric cancer,so that p53 and P gp gene products immunostaining assay at the early stage of gastric cancer may be of great clinical significance.
