摘  要:从烟叶叶面分离到产果胶酶真菌 18株 ,在此基础上 ,进行液体培养 ,测定酶活 ,得到一株酶活性较高的黄曲霉DPE - 0 0 5。对该菌株产酶培养基的碳、氮源进行正交法研究 ,正交实验的结果表明 ,影响该菌产酶活性的因素依次为A(麸皮 ) >B(乳糖 ) >C(果胶 ) >D(硫酸铵 ) ,其最佳组合为A2B3C3D1。最适产酶条件为 :麸皮 4 0 % ,果胶 0 3% ,乳糖 1 5 % ,(NH4 ) 2 SO4 0 5 % ,KH2 PO4 0 2 5 % ,MgSO4 ·7H2 O 0 0 5 % ,NaNO30 0 2 % ,FeSO4 ·7H2 O 0 0 0 1% ,起始pH 6 0 ,在 2 8℃摇床培养 7d产酶量达到最高。以玉溪B3F烟叶为材料 ,施加DPE - 0 0 5菌株所产酶液 ,在 5 0℃贮存 12h。经化学成分检测 ,结果表明 ,果胶质降低了 18 15 %。strains of funguses were isolated from tobacco leaves A strain No DPE-005 with high pectinase activity was obtained by repeated screening The enzyme-production conditions of carbon and nitrogen of it were studied through the method of experiments of orthogonal matrix The experimental result showed that the influential extent of affecting factors to enzyme-production of No DPE-005 is:A(bran)>B(lactose)>C(pectin)>D((NH 4) 2SO 4) The best combination of enzyme-production is A2B3C3D1 The optimum medium of enzyme-production are as follows:4 0%bran,0 3%pectin,1 5%lactose,0 5%(NH 4) 2SO 4,0 25% KH 2PO 4,0 05% MgSO 4·7H 2O,0 02% NaNO 3,0 001% FeSO 4·7H 2O,pH 6 0,After seven day's shake culture at 28℃,the maximum activity of enzyme was produced Tobacco(Yuxi B3F) was added the enzyme solution produced by DPE-005 into and stored in 50℃ for 12hours The content of pectin was reduced by 18 15%
