摘  要:目的:观察肝癌形成过程中MMP-2mRNA表达的动态变化,探索明胶酶在肝癌生长浸润中的作用,应用明胶酶抑制剂Batimastat(BB-94)进行治疗,观察其对明胶酶mRNA表达的影响。方法:应用RT-PCR法对大鼠肝癌形成过程中各期MMP-2mRNA表达情况进行观察,并与Batimastat治疗组和正常对照组进行统计分析。结果:诱癌过程中早期肝硬化期MMP-2mRNA表达成缓慢增高趋势,至癌变期表达明显较增高,并呈一平台,诱癌不同时期应用BB-94对MMP-2mRNA相对表达值无影响。结论:诱癌过程中MMP-2mRNA表达持续升高,BB-94治疗对MMP-2mRNA的表达没有影响。AIM: To study the dynamic changes of matrix rnetalloproteinase-2 mRNA in liver tissue during the experimental hepatocarcinogenesis and the effect of Batimastat on it. METHODS: Hepatocellular carcinoma was induced with the administration of diethy Initrosoamine(DENA) in rats. BB-94 was intraperitoneally injected to treat the experimental models. Reverse transcription polymerase chain reaction was used for quantitative analysis of MMP-2 mRNA expression level during the induction and therapy. RESULTS: The expression of MMP-2mRNA was increased throughout the hepatocardnogenesis, and reach had its maximum plateau in the early hepatocarcinogenesis stage. BB-94 no effect on MMP-2mRNA expression. CONCLUSION: MMP-2mRNA expression is increasing during hepatocarinogenesis, and BB-94 has no effect on MMP-2mRNA expression.
