摘  要:目的 :确定内毒素 (LPS)诱导巨噬细胞产生肿瘤坏死因子 (TNF)的适宜用量 ,建立内毒素感染的细胞模型。方法 :选用不同剂量的LPS ,刺激大鼠腹腔巨噬细胞 ,分别于不同的刺激时间收集细胞培养上清。TNF细胞毒效应用L929细胞和WEHI-164细胞检测,TNF -α浓度用ELISA法检测。结果 :当LPS使用浓度为0.1μg/ml至5μg/ml时 ,其诱导产生的TNF细胞毒效应有逐渐增加的趋势 ,超过此剂量时 ,TNF细胞毒效应反而降低 ;巨噬细胞TNF的分泌量也随LPS剂量的增加而减少。结论 :LPS在一定的浓度范围内 ,巨噬细胞培养上清中TNF含量明显增多 。Objective: To determine the suitable amount of lipopolysaccharide (LPS), which is used to induce the macrophages to produce tumor necrosis factor (TNF), setting up a cellular model of endotoxin infection.Methods:The rat abdominal cavity macrophages were stimulated by LPS in different dose, and then, in different stimulation time, collected cell culture supernatant. Determination of the TNF cytotoxic effect were performed in L929 and WEHI-164 cell line,the concentration of the TNF-α was determined by ELISA.Results:When the LPS doses was in a range from 0.1 μg /ml to 5 μg /ml,the TNF cytotoxic effect increased gradually; over this doses, the TNF cytotoxic effect fell.And the product of the TNF in the macrophages decreased with the LPS increase.Conclusion:Within the certain concentration range, LPS may lead to a significant increase in the TNF content in the macrophages culture supernatant, but excessive the LPS may decrease the TNF secretion.
