摘  要:目的 探讨可利新对肝硬化大鼠门脉压力、胃粘膜血流量及血管活性物质一氧化氮 (NO)、内皮素 (ET)、胰高糖素 (Glucagon)及血管紧张素Ⅱ (AngⅡ )的影响。方法  6 6只雄性SD大鼠随机分为 2部分 ,每部分再分为胆管结扎组、假手术组和对照组 ,前 2组尾静脉注射可利新 (0 2mg/kg) ,对照组注射同等剂量生理盐水。第 1部分采用多导生理记录仪测定门脉压力和胃粘膜血流量 ,第 2部分分别用Griess法测定血浆NO含量、放免法测定血浆ET、Glucagon和AngⅡ含量。结果 用药后门脉压力明显降低 ,胃粘膜血流量减少 ;血管活性物质的水平亦有明显降低 ,与用药前相比其差异具有显著意义。结论 可利新不仅通过降低门脉压和胃粘膜血流量来治疗肝硬化及其并发症 。Objective To investigate the effects of Glypressin on portal vein pressure (PVP), gastric mucosal blood flow and vaso-active substances such as NO, ET, glucagon or angiotensin Ⅱ (Ang Ⅱ) in cirrhotic rats. Methods Sixty-six male SD rats were divided into in two parts and each was divided into three groups: common bile duct ligation (CBDL) group, sham-operated (sham) group and control group. The model of cirrhotic rats was made by CBDL. The rats in the sham group and control group were only subjected to incision of abdominal cavity. The rats in the CBDL and sham group were given with glypressin (0.2 mg/kg), while those in the control group were given with the same volume of normal saline. The PVP and gastric mucosal blood flow were measured by polygraph. Plasma NO content and contents of ET, glucagon and ang Ⅱ were determined by Griess method and radioimmunoassay, respectively. Results After administration of glypressin, the PVP, gastric mucosal blood flow and the levels of the vaso-active substances were decreased significantly as compared with those before medication. Conclusion Glypressin can be used to treat liver cirrhosis and complications by decreasing not only PVP and gastric mucosal blood flow, but also by vaso-active substances.
