P-选择素单抗抗唾液腺腺样囊性癌细胞裸鼠肺粘附的研究

摘　　要：目的观察P-选择素单克隆抗体(MonoantibodytoP-selectin,PS1)对腺样囊性癌肺高转移细胞肺粘附的抑制作用。方法裸鼠尾静脉注射P-选择素单抗后,经尾静脉注射氚标胸腺嘧啶核苷(3HTdR)标记的ACC-M细胞,分别检测细胞注射后2h、6h、18h的单位重量肺、肝组织每分钟核素脉冲数(CPM)。结果空白对照组单位重量肺组织在2h、6h、18h的CPM值分别是1875.75±11.53、1644.50±27.20和1466.50±125.95;P-选择素单抗组肺组织2h、6h、18h的CPM值分别是941.50±68.47、875.00±47.06和787.25±65.38。相同时间P-选择素单抗组肺组织CPM值明显低于对照组,两组间有显著性差异(P<0.001)。空白对照组单位重量肝组织的CPM值与P-选择素单抗组相比,2h、6h、18h各相同时间两组间无显著性差异(P>0.05)。结论P-选择素单抗静脉应用后,可减少肺组织内ACC-M细胞的粘附数量,但对相同时间肝组织内ACC-M细胞数量无显著影响。Objective To investigate the effect of intravenous administration of monoantibody to P-selectin(PS1)on the lung-metastasis characteristic of highly lung metastatic adenoid cystic carcinoma cell strain(ACC-M).Methods 3 HTdR labeled ACC-M cells were injected intravenously through tail vein after using monoantibody to P-selectin.4mice of each group were killed at2h,6h and18h after infusion,respectively.The relative radioactivity(CPM)in the lung and liver was detected.Results 3 H-activity per gram in lung of mAb group was lower than in control group.There was significant difference between the two groups(P<0.001).There was no difference between the two groups of liver tissues(P>0.05).Conclusions The results of this study suggested that the highly lung metastasis characteristic of ACC-M may be partially inhibited by the use of monoantibody to the P-selectin.
