摘 要:目的 :观察IL - 13对急性肾缺血再灌注时IL - 1β表达的影响。 方法 :Wistar雄性大鼠 5 7只 ,随机分为 8组 :正常组 (normal) ;假手术组 (sham) ;缺血组 :(I)缺血再灌注组 (I/R) ;治疗对照组 - 1(C - 1) ;治疗对照组 - 2 (C- 2 ) ;治疗组 - 1(T - 1)和治疗组 - 2 (T - 2 )。阻断大鼠双侧肾脏血流 4 5min再灌注 2 4h建立急性肾缺血再灌注模型 ;治疗组分别于阻断血流前、后分别从双侧肾动脉开口注射入 1 5 μg/ 5 0gbw鼠重组白细胞介素 13(rmIL - 13) ;检测各组大鼠IL - 1β血清水平和肾脏表达 ,以及肾功能和肾脏病理。 结果 :(1)治疗组肾脏IL - 1β基因 (TtoC :P <0 0 1)和蛋白表达 (T - 1toC - 1:P <0 0 1;T - 2toC - 2 :P <0 0 5 )明显减少 ,血清IL - 1β水平明显下降 [C - 1toT - 1:(2 7 13± 5 5 1)ng/Lto (14 0 5± 3 82 )ng/L ,P <0 0 1;C - 2toT - 2 :(2 9 5 2± 5 84 )ng/Lto (14 2 7± 3 5 2 )ng/L ,P <0 0 1];(2 )肾功能障碍和肾组织病理变化明显减轻 ,肾小管损害评分减少 (C - 1toT - 1:4 5 2 0± 8 6 4to 2 1 0 5± 8 82 ,P <0 0 1;C - 2toT - 2 :4 2 2 5± 11 15to 2 3 2 5± 7 31,P <0 0 1) ;(3)血清IL - 1β水平与BUN、Cr成正相关 (r=0 70 8,P <0 0 1;r=0 770 ,P <0 0AIM: To investigate the effects of IL-13 on expression of IL-1β in acute renal ischemia/reperfusion injury. METHODS: Fifty-seven male Wistar rats were randomly divided into 8 group: normal group,sham operation group, ischemia group, ischemia/reperfusion injury group(I/R), normal saline(NS)-treated group 1(C-1), NS-treated group 2(C-2),IL-13-treated group1(T-1)and IL-13-treated group 2(T-2).Rats were subjected to 45 min bilateral renal ischemia followed by reperfusion. rmIL-13 (1.5 μg/50 g body weight )was injected into the renal arteries through the abdominal aorta before ischemia(T-1) or immediately afterischemia(T-2).The serum level of IL-1β and the renal expression of IL-1β were determined in each group at 24 h post-ischemia. In addition, BUN,Cr and renal histology were also measured. RESULTS: (1)The serum level of IL-1β [C-1to T-1: (27.13±5.51) ng/L to (14.05±3.82) ng/L, P< 0.01;C-2 to T-2: (29.52±5.84) ng/L to (14.27±3.52) ng/L, P< 0.01], gene expression and protein production of IL-1β in kidney decreased markedly in IL-13-treated groups.(2)Renal function and histology were significantly improved in IL-13-treated groups, renal injury scores decreased significantly.(3)A positive correlation were found between the serum level of IL-1β and BUN,SCr( r= 0 708, P< 0 01; r= 0 770, P< 0 01).CONCLUSION: These data suggest that IL-13 inhibit the expression of IL-1β and improve function and histology of kidney in acute renal ischemia/reperfusion injury.
