放疗对涎腺腺样囊性癌细胞膜型基质金属蛋白酶-1表达的影响

摘  要:①目的 研究放疗对人涎腺腺样囊性癌细胞株ACC M膜型基质金属蛋白酶 1 (MT1 MMP)表达的影响。②方法 以60 Co为放射源 ,对放疗组腺样囊性癌细胞分别给予 2~ 8Gy不同剂量的单次照射 ,2 4h后利用逆转录聚合酶链反应 (RT PCR)和流式细胞术对其MT1 MMPmRNA和蛋白表达进行检测。对照组细胞不予放疗处理。③结果 不同剂量放疗组细胞MT1 MMPmRNA和蛋白表达均低于对照组 ,差异有显著性 (F =1 3.76、2 5 7.36 ,q =6 .71~ 39.0 5 ,P <0 .0 1 )。 6Gy和 8Gy剂量放疗组MT1 MMPmRNA表达低于 2Gy和 4Gy组 (q =1 4 .2 8~ 2 0 .6 3,P <0 .0 1 ) ,不同剂量放疗组细胞MT1 MMP蛋白表达无显著性差异 (q =0 .5 1~ 2 .5 3,P >0 .0 5 )。④结论 放疗能抑制人涎腺腺样囊性癌细胞株ACC M细胞MT1Objective\ To investigate the effect of radiotherapy on the membrane type matrix metalloproteinase 1 (MT1 MMP) expression of human salivary gland adenoid cystic carcinoma cell line ACC M in vitro. \ Methods\ Reverse transcriptase polymerase chain reaction (RT PCR) was applied to study the mRNA expression of MT1 MMP on ACC M cells at 24 h after irradiation with graded single dose of 2-8 Gy gamma rays, a flow cytometric analysis was performed to detect the MT1 MMP expression at protein level. The control cells didnt receive irradiation. \ Results\ The study revealed that the mRNA and protein expression of MT1 MMP was suppressed on ACC M cells at 24 h after different dose irradiation ( F=13.76,257.36, q=6.71- 39.05 , P <0.01). Irradiation with doses of 6 Gy and 8 Gy suppressed the MT1 MMP mRNA expression significantly compared with 2 Gy and 4 Gy ( q=14.28-20.63, P <0.01), there was no difference in MT1 MMP protein expression between the irradiated groups ( q=0.51-2.53, P >0.05). \ Conclusion\ Radiotherapy suppressed MT1 MMP expression on ACC M cells in vitro.
