摘 要:目的 :研究脆性组氨酸三联体 (fragile histidine triad,FHIT)基因在电离辐射损伤和辐射致癌早期过程中的作用。方法 :将 4 0只 BAL B/ c小鼠随机分成对照组和 1.8、3.6、5 .4、7.2 Gy照射组共 5组 ,每组 8只动物。照射后 1个月用乙醚麻醉小鼠 ,心脏抽血 ,再解剖取胸腺及骨髓细胞 ,抽提其总 RNA。采用巢式 RT- PCR方法及 PCR产物直接测序法检测 FHIT基因的表达情况。 结果 :对照组血液、胸腺及骨髓均未出现异常 FHIT转录本的表达 ,不同剂量照射组中血液、胸腺及骨髓均有部分样品出现相对分子质量较小的异常 FHIT转录本的表达。测序证实异常转录本缺失 2个或 2个以上的 FHIT外显子。结论 :电离辐射可引起 FHIT基因的缺失 。Objective:To investigate the role of fragile histidine triad(FHIT) gene in irradiation injury and irradiation carcinogenesis.Methods:Forty BALB/c mice were divided into 1.8,3.6,5.4,7.2 Gy irradiation groups and control group at random.Mice were killed a month after they were irradiated.Total RNAs were extracted from blood,thymus glands and bone marrow.The expression of FHIT gene was studied by means of nested RT PCR and DNA sequencing.Results:Abnormal transcripts of FHIT gene were not observed in blood,thymus and bone marrow of control group.Aberrant transcripts of FHIT gene were detected in blood,thymus and bone marrow in different irradiation dose groups, and the deletion of FHIT gene was confirmed by DNA sequencing.Conclusion:The results suggest irradiation can cause the deletion of FHIT gene, FHIT gene may play an important role in radiation injury and radiation carcinogenesis.
