摘  要:探讨端粒长度与端粒酶活性在人鼻咽癌细胞CNE 2L2恶性行为改变前后的变化 ,建立研究恶性行为改变与端粒长度与端粒酶活性间关系的细胞模型。与 6A8α 甘露糖苷酶表达正常的CNE 2L2细胞 (野生型细胞W ,转导空载体的细胞M及转导无关DNA片段的细胞S)相比 ,6A8α 甘露糖苷酶表达低下的细胞 (AS)接种裸鼠皮下后的肿瘤性生长受抑。用TeloTAGGGTelomereLengthAssayKit及TelomerasePCRELISAKit分别测定端粒长度及端粒酶活性 ,用RT PCR方法分析端粒重复序列结合因子 (TRF)的转录水平。见AS细胞的端粒明显缩短 (6 78Kb ,W细胞为8 4 0Kb ,M细胞为 8 34kb ,S细胞为 9 5 6kb) ,但端粒酶活性及端粒重复序列结合因子 1和 2 (TRF 1和 2 )的转录水平未见改变。实验表明 ,恶性行为降低的CNE 2L2细胞的端粒变短 ,但与端粒酶活性及TRF 1 2无关 ,提示在CNE 2L2细胞中可能存在着端粒酶及TRF 1 2以外的调节端粒长度的机制。Changes of telomere length and telomerase activities in CNE 2L2 cells with malignancy reduction were analyzed and a cell model for exploring relationships between changes of malignancy and telomere length and telomerase activities was developed. In contrast to the growth of tumors from the CNE 2L2 cells with normal expression of 6A8 α mannosidase (W, the wild type cell; M, mock transduced cell; or S, an unrelated DNA transduced cell), that of the tumor from the cells with reduction of 6A8 α mannosidase expression (AS) inoculated into nude mice was greatly inhibited. Telomere length and telomerase activity were assayed using Telo TAGGG Telomere Length Assay kit and Telomerase PCR ELISA kit, respectively. Telomere repeat binding fragment 1 and 2 (TRF1 and 2) mRNA expression were detected by means of RT PCR. Telomere length loss was found in AS cell (6.78kb versus 8.40kb in W cell, 8.34kb in M cell and 9.56kb in S cell). However, no change in telomerase activity and TRF 1 and 2 mRNA transcription was observed in AS cell. The results suggest the existence of alternative mechanism(s) responsible for the telomere length loss in CNE 2L2 cell with malignancy reduction. A cell model for studying relationships between changes of malignancy and telomere length and telomerase activities has been developed.
