摘 要:目的 探讨视网膜消化铺片联合免疫荧光染色技术的可行性和应用价值。方法 健康成年雄性 Wistar大鼠 12只 ,随机分成正常对照组和链尿佐菌素 (Streptozotocin,STZ)诱导的糖尿病 3个月组各 6只 ;取每只大鼠左眼作视网膜胰酶消化铺片 ,并在铺片上进行血管细胞粘附分子 - 1(VCAM- 1)单克隆抗体免疫荧光组织化学研究。结果 正常对照组视网膜血管走行规则 ,未见阳性细胞表达 ;糖尿病 3个月组视网膜血管走行尚规则 ,并可见血管腔节段性膨大 ,血管壁见阳性细胞表达。结论 视网膜血管消化铺片联合免疫荧光组织化学技术是可行的 ,为视网膜血管性病变的研究提供了重要的途径。Objective To explore the feasibility and applicative value of retinal digest preparations combined with immunofluorescence in the retinal vascular diseases.Method 12 healthy adult male Wistar rats were randomly divided into normal control group and 3 months of diabetes (STZ) group. Retinal digest preparations were made and immunofluorescence of the vascular cell adhesion molecule 1 (VCAM 1) was detected with monoclonal antibody.Results The retinal vessels arranged regularly and there were not positive cells in the control group;It was found that the retinal microvessels expanded partly and the positive cells of VCAM 1 expressed significantly in the 3 months of diabete group.Conclusion Retinal digest preparations combined with immunofluorescence. was feasible and provided an important method for the studies of retinal vascular diseases.
