鼻咽癌细胞株CNE1转染EB病毒潜伏膜蛋白1(LMP1)后对放射敏感性的影响

摘　　要：目的 :研究 EB病毒 LMP1对鼻咽癌高分化细胞株 CNE1放射敏感性的影响。方法 :用电转染技术将带有 EB病毒 LMP1基因的真核表达质粒导入鼻咽癌高分化细胞株 CNE1 ,用免疫组化法及 Western blot检测 LMP1的表达 ;LMP1对 CNE1细胞放射敏感性的影响采用6 0钴治疗仪照射后集落形成实验进行观察。结果 :CNE1细胞株转染 LMP1基因后 LMP1蛋白呈阳性表达 ,对照放射敏感性比未转染株的细胞存活率低 (P<0 .0 1 )。结论 :LMP1能增加鼻咽癌细胞的放射敏感性 ,这种作用可能与 LMP1的促细胞转化 ,抑分化有关。Objective:To study the effect of EBV latent membrane protein 1(LMP1)gene on the radiosensitivity of high differentiated nasopharyngeal carcinoma cell line CNE1 Methods:EBV LMP1 gene eukaryotic expression plasmid was transfected into CNE1 cells with electroporation technique LMP1 expression in the transfectants was detected by immunohistochemical methods and Western blot The livability of cells after 60 Co irradiation was measured by clone forming assay Results:The transfectants expressed LMP1 The cell livability after 60 Co radiation in transfected cells was lower than that in untransfected cells Conclusion:LMP1 gene could increase the radiosensitivity of nasopharyngeal carcinoma,and the effect might be related to its ability of facilitating cell transformation and inhibiting cell differentiation
